    ÄKTA oligopilot plus is a flexible and fully-automated DNA/RNA oligonucleotide synthesizer designed for use in research and for process development. ÄKTA oligopilot plus is designed to meet the needs of various oligonucleotide chemistries and therapeutics (RNAi, siRNA, aptamer, antisense, immunostimulating CpG motifs, Exon skipping) enabling cost-efficient synthesis of high-quality oligonucleotides.

    Flow-through reactor technology

    The system employs flow-through reactor technology, which results in significantly reduced reagent consumption, enables exact control of reaction rates and contact times, and allows smooth scale-up.

    Column re-circulation

    ÄKTA oligopilot plus features column re-circulation, an important feature for extending the time of coupling reactions, of significant impact when coupling RNA or different specific modified amidites. ÄKTA oligopilot plus is available in two different configurations: ÄKTA oligopilot plus 10 for synthesis in the 10 - 50 μmol range and ÄKTA oligopilot plus 100 for synthesis in the 50 μmol - 9 mmol range.

    Fastest possible column wash

    Pressure controlled flow and conditional method programming based on UV signals allows the fastest possible column wash between synthesis steps and reduces synthesis cycle time. The batch operation and documentation are in accordance with the requirements of current good manufacturing practice (cGMP), good laboratory practice (GLP), and is in full compliance with 21 CFR Part 11.

    Compatible with a range of column reactors

    ÄKTA oligopilot plus is compatible with a range of column reactors ranging from fixed column volume 1.2 mL – 48 mL to larger, adjustable, column volumes of FineLINE 35 oligo (scales from 250 μmol - 4 mmol) and the Adjustable column (<9 mmol). Monomer sliders and bottle caps have been developed for use with ÄKTA oligopilot plus. The sliders fit on existing mounting rails on the instrument side. Bottle caps have one gas connection, and three reagent connections that fit all different capillaries used with the system.

    About UNICORN control software

    ÄKTA oligopilot plus is controlled via an external computer using UNICORN software especially designed for production needs. Features such as a sequence editor for up to 200 bases, password protection for different access levels, lot number registration, and full synthesis documentation are available. Combining ÄKTA oligopilot plus extensive monitoring capabilities with UNICORN allows a PAT (Process Analytical Technology) approach. Online trityl monitoring and coupling efficiency display provide security during synthesis, shutting down the system if coupling efficiency drops below a pre-determined threshold.
